Always take Olaparib exactly as your doctor or pharmacist tells you. Olaparib tablets should be swallowed whole with water. Do not chew, crush, dissolve, or split the tablets, as this may affect how quickly the medicine is absorbed into your body. You may take it with or without food. The recommended dosing frequency is twice daily: once in the morning and once in the evening. The usual recommended dose is 300mg (i.e., two 150mg tablets) twice a day, for a total of 4 tablets each day. Be sure to take the full recommended dose every day, continue taking it for as long as your doctor advises, and do not stop on your own.
Your doctor may adjust your dose in the following three situations: First, if you have kidney problems, your doctor may ask you to take 200mg (i.e., two 100mg tablets) twice a day, also a total of 4 tablets per day. Second, if you are taking certain medicines that may affect how Olaparib works (see the interactions section above for details), your doctor will adjust the dose as needed. Third, if you experience certain side effects while taking the medicine (e.g., severe reduction in blood cells), your doctor may temporarily reduce your dose, interrupt treatment for a short period, or permanently stop treatment. Any dose adjustment must be done under your doctor's guidance.
If you forget a dose, do not panic; just take your next dose at the next scheduled time as normal. Do not take a double dose (i.e., two doses at once) to make up for the missed dose. If you think you may have taken an overdose of Olaparib, contact your doctor immediately or go to the nearest hospital. Regarding contraception and pregnancy: Female patients must use two effective methods of contraception during treatment and for 6 months after the last dose, and must undergo regular pregnancy testing before, during, and for 6 months after treatment. Male patients must use condoms during treatment and for 3 months after the last dose, and must not donate sperm. Breastfeeding women must not breastfeed during treatment and for 1 month after the last dose.
