Repotrectinib may cause serious side effects, among which central nervous system issues require particular attention. During treatment, symptoms such as dizziness, vertigo, mood changes (e.g., anxiety, irritability, depression), balance or coordination problems, confusion or forgetfulness, hallucinations (seeing or hearing things that are not there), as well as issues with attention, memory, and sleep may occur. Warning signs of liver problems include: yellowing of the skin or whites of the eyes, dark or tea-colored urine, light-colored stools, loss of appetite, nausea and vomiting, and pain in the upper right side of the abdomen. If any of these symptoms occur, contact your doctor immediately. At the same time, repotrectinib may also cause muscle problems. Blood tests are also required before treatment and early in the treatment period. Manifestations of muscle problems include: unexplained or persistent muscle pain, muscle tenderness, and unexplained weakness. Be sure to inform your healthcare team if you experience any new or worsening muscle symptoms.

Hyperuricemia and Risk of Fractures

Repotrectinib may cause abnormally high levels of uric acid in the blood, known as hyperuricemia. Your doctor will monitor uric acid levels through blood tests before and after treatment, and may prescribe uric acid-lowering medications if necessary. Symptoms of hyperuricemia include: redness, heat, tenderness, or swelling in joints (especially the big toe), abdominal or flank pain, significantly reduced urine output or no urine output, nausea and vomiting, and urine that is pink, brown, or bloody. Seek immediate medical attention if these symptoms occur. Furthermore, repotrectinib may increase the risk of fractures. Fractures can occur after a fall or injury, or sometimes without an obvious cause. If you experience unexplained pain, changes in mobility, or feel that your bones are abnormal during treatment, contact your healthcare provider immediately. Early detection and management of these side effects help ensure the safety of treatment.
